Hemp seed oil is well known for its nutraceutical, cosmetic and pharmaceutical properties due to a perfectly balanced content of omega 3 and omega 6 polyunsaturated fatty acids. Its importance for human health is reflected by the success on the market of organic goods in recent years. However, it is of utmost importance to consider that its healthy properties are strictly related to its chemical composition, which varies depending not only on the manufacturing method, but also on the hemp variety employed. In the present work, we analyzed the chemical profile of ten commercially available organic hemp seed oils. Their cannabinoid profile was evaluated by a liquid chromatography method coupled to high-resolution mass spectrometry. Besides tetrahydrocannabinol and cannabidiol, other 30 cannabinoids were identified for the first time in hemp seed oil. The results obtained were processed according to an untargeted metabolomics approach. The multivariate statistical analysis showed highly significant differences in the chemical composition and, in particular, in the cannabinoid content of the hemp oils under investigation.

The influence of the single components of Ferula hermonis extract on sexual behavior was studied in male rats. Sexually potent and sluggish/impotent animals were orally treated acutely (2.5 mg/kg) and subchronically (0.25 mg/kg/day for 10 days) with ferutinin, teferdin and teferin. Ferutinin alone acutely administered in potent rats was able to reduce mount and intromission latencies, while in sluggish/impotent animals, it induced the same effects and additionally shortened the ejaculation latency, as teferdin did. Both substances increased testosterone levels in rats. Unlike teferdin, ferutinin subchronically administered in potent rats negatively affected appetitive and consummatory sexual behavior, reducing also testosterone serum levels. In conclusion, if repetitively administered, ferutinin was able to stimulate sexual behavior after acute ingestion, but exerted a negative influence on the sexual capacity of potent male rats, whereas teferdin only improved copulatory performance of sluggish/impotent animals.

The thiophene-, benzothiazole- and pyridine-thioaryloxyacids analogues of clofibric acid were synthesized and their antiplatelet activity was screened. Some compounds exhibited antiaggregating properties. The platelet-related haemostasis was measured on a PFA-100 (R) analyzer using bull blood.


2003 - Opposite effect of acute and subchronic treatments with Ferula hermonis on copulatory behavior of male rats [Articolo su rivista]

Sexually potent and sluggish/impotent male rats were orally treated with an extract of Ferula hermonis (30 and 60 mg/kg). The acute administration stimulated sexual motivation in potent rats and improved copulatory performance in sluggish/impotent rats. This last effect was elicited only by the higher dose, which, in parallel, increased serum testosterone levels in rats. On the contrary, when the extract was subchronically administered (10 days) a marked reduction in the percentage of rats achieving ejaculation was detected, together with a general impairment of the copulatory pattern. Furthermore, the repeated administration of the extract (6 mg/kg/day for 10 days) resulted in a significant reduction of testosterone levels in comparison with controls. The present results discourage a repeated assumption of F. hermonis, while suggesting its acute administration to improve the performance in sexual dysfunctions.

The chiral analogues of gemfibrozil 5-(2,5-dimethylphenoxy)-2-methylpentanoic acid and 5-(2,5-dimethylphenoxy)-2-ethylpentanoic acid were synthesized in optically active form using (S)-4-(1-methylethyl)-2-oxazolidinone as chiral auxiliary. All compounds inhibit human platelet aggregation. From these data, one can surmise that all tested compounds and gemfibrozil act at the platelet level with different mechanism than that of ASA, even if with a different potency. © 2002 Elsevier Science Ltd. This paper reports the use of liquid chromatography for the separation and determination of the major cannabinoids extracted from hashish samples. The direct coupling to the mass spectrometer enables the selective identification both of neutral and acidic cannabinoids. The developed method does not require any preliminary derivatization and should, therefore, be of interest in forensic analysis for simple and unequivocal determination of hashish constituents.

An analytical procedure has been developed for the determination of zinc pyrithione in antidandruff formulations. Zinc pyrithione was converted into a stable copper complex and then analysed by reversed-phase HPLC. The proposed method allows the separation of the analyte from related pyrithiones and therefore is able to verify the compliance of cosmetic preparations with current legislation.

A rapid and simple HPLC method for the identification and determination of dihydroxyacetone, a tanning agent widely used in a variety of preparations, is presented. The method consists of the analysis of the formulated products after a derivatization reaction which enhances the response and the detection limit of the chromatographic analysis. The characterization and the elucidation of the spacial geometry of the synthesized derivative are described.

The binary phase diagram of [S,S(+)]-ethambutol dihydrochloride, a chemotherapeutic drug, and its [R,R(-)]-enantiomer, has been determined by IR spectroscopy, X-ray podwer diffraction and differential scanning calorimetry, the equimolecular mixture of [S,S(+)]/[R,R(-)] corresponds to a racemic compound. The infrared spectra of enantiomers and racemic compound differ especially in the finger print region. The solid-solid transition and fusion enthalpies and entropies for the optically active compounds and the corresponding racemic compound have been determined. The stability of the racemic compound has been evaluated on the basis of free energy of formation. In this paper the results of calorimetric and spectroscopic analyses for the determination of enantiomeric purity of ethambutol dihydrochloride have been reported.

This paper reports a RP-HPLC method for the determination in topics of azelaic acid, a keratolytic and anti-comedogenic agent widely used in the treatment of all types of acne. A derivatization step was needed prior to chromatographic analysis because the analyte is lacking in chromophore. A sample clean-up procedure by solid-phase extraction was also developed to analyse azelaic acid in complex matrices, such as pharmaceutical and cosmetic formulations.

A direct liquid chromatographic method with UV detection has been developed for the determination of ethambutol dihydrochloride, a chemotherapeutic drug lacking in chromophoric groups. It allows the simultaneous separation of [S,S(+)]-ethambutol from meso-[S,R(+/-)]-form and its synthetic precursor [S(+)]-2-amino-1-butanol, although it is not stereospecific for the separation of the [R,R(-)]-enantiomer. Important factors affecting the selectivity and efficiency of separation (CuSO4 concentration, temperature and addition of an organic modifier in the mobile phase) have been studied. Moreover the proposed method allows the quantitative determination of ethambutol dihydrochloride in pharmaceutical formulations in order to verify the agreement with the claimed content for the drug.

The proposed method allows the routine detection of these preservatives in different cosmetic formulations with a view for verifying their complicance with Italian legislation.
